Assay Genie offers a colorimetric, solid-phase enzyme-linked immunosorbent assay (ELISA) based on the sandwich principle for quantitative analysis of free daratumumab (Darzalex®) in serum and plasma samples. Standards and samples are incubated in microtiter plate wells coated with a daratumumab reactant. Following washing, a horse radish peroxidase-conjugated probe is added, and bound enzymatic activity is detected using tetramethylbenzidine chromogen substrate before the reaction is terminated with an acidic stop solution.

The developed colour is proportional to the amount of daratumumab in the sample or standard, allowing results to be determined using the standard curve. Application details list serum and plasma as sample types, with a required volume of “10 µL” and total time of “70 Minutes”. The stated detection limit is “30 ng/mL”, and spike recovery is “85-115%”.
