Description

Assay Genie's phospho-specific Cell-Based ELISA Kit is configured to detect DAPP1 phosphorylated at Tyr139, supporting research into immune cell signaling and downstream signal transduction. The colorimetric readout at 450 nm uses an indirect ELISA format in which HRP-conjugated secondary antibodies catalyze the substrate reaction, and the assay is high-throughput and lysate-free, allowing measurement of relative DAPP1 phosphorylation in cultured cells as well as screening for effects of treatments, inhibitors such as siRNA, or activators.

Reactivity covers human and mouse samples, and the kit is designed for use with adherent cells, with a stated dynamic range of more than 5,000 cells. An anti-GAPDH monoclonal antibody is included as an internal positive control for normalization of absorbance values, and a Crystal Violet whole-cell staining step can be used to normalize readings to cell density and adjust for plating differences.

DAPP1 is described as an adaptor protein that regulates antigen receptor signaling downstream of phosphatidylinositol 3-kinase, with phosphorylation at Tyr139 reported as critical for its function in mediating cellular responses to external stimuli. Dysregulation of DAPP1 signaling has been implicated in cancer, autoimmune disorders, and inflammatory conditions, making the phospho-Tyr139 target relevant to disease pathogenesis studies and drug development research.
