Description

Assay Genie's DAPP1 Colorimetric Cell-Based ELISA Kit is designed for the qualitative measurement of DAPP1 protein levels in cell lysates and culture supernatants. The kit employs an indirect ELISA format in which DAPP1-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. Detection is read at 450 nm.

Reactive with human and mouse samples, the assay is intended for adherent cells and is reported to have a dynamic range of > 5000 Cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining step is provided for cell density determination. The kit is supplied with cell-culture-treated microplates, 100x anti-phospho target and anti-target antibodies, HRP-conjugated anti-rabbit and anti-mouse IgG antibodies, buffers, ready-to-use substrate, stop solution, and adhesive plate seals. It is stored at 4 °C with a stated stability of 6 months.

DAPP1 (dual adapter for phosphotyrosine and 3-phosphotyrosine and 3-phosphoinositide, also known as BAM32) is an adaptor protein reported to regulate antigen receptor signaling downstream of phosphatidylinositol 3-kinase, with described roles in cell growth, proliferation, and survival. Dysregulation of DAPP1 has been implicated in cancer, autoimmune disorders, and inflammatory conditions.
