Description

Assay Genie offers a colorimetric, cell-based ELISA kit designed for the quantitative detection of DAPK3 (Death-Associated Protein Kinase 3) phosphorylated at threonine 265. Detection is read at 450 nm, and the kit is reactive with human, mouse, and rat samples, with a dynamic range of > 5000 cells. The assay is intended for use with adherent cell samples and provides a lysate-free workflow.

According to the manufacturer, the kit is a convenient, high-throughput, and sensitive assay that detects DAPK3 protein phosphorylation and expression profile directly in cultured cells. It uses an indirect ELISA format, in which DAPK3 (Phospho-Thr265) is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies, with a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method is supplied so that absorbance values can subsequently be normalised to cell density.

DAPK3 is a serine/threonine kinase implicated in the regulation of apoptosis, autophagy, transcription, actin cytoskeleton reorganisation, cell motility, smooth muscle contraction, and cytokinesis. Phosphorylation at threonine 265 has been linked to regulation of its enzymatic activity and cellular functions, making the phospho-Thr265 form a relevant target for studies in cancer, neurodegeneration, and inflammation. Associated database entries include Gene ID 1613 and UniProt ID O43293, with a reported molecular weight of ~ 52kDa.
