Description

This in-cell, lysate-free ELISA kit from Assay Genie is designed to detect and quantify Cytochrome P450 7B1 protein expression directly within cultured cells. The assay employs an indirect ELISA format in which target-specific primary antibodies capture the Cytochrome P450 7B1 protein, while HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition, with the plate read at 450 nm.

Designed for use with human adherent cells, the kit supports qualitative measurement of Cytochrome P450 7B1 levels and is suitable for screening the effects that various treatments, inhibitors (e.g. siRNA or chemicals), or activators have on Cytochrome P450 7B1. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values. Following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts.

Cytochrome P450 7B1 (gene symbol CYP7B1, UniProt ID: O75881) is an endoplasmic reticulum membrane-bound oxysterol 7-alpha-hydroxylase involved in the metabolism of cholesterol and bile acids, and plays a critical role in the regulation of lipid homeostasis. Dysregulation of this enzyme has been linked to various metabolic disorders, making it a valuable biomarker for studying these conditions and developing targeted therapies. The kit has a dynamic range of > 5000 Cells and is supplied in a 96-well microplate format, with a stability of 6 months at 4°C.
