Description

From Assay Genie, this Cell-Based ELISA Kit provides a colorimetric method (read at 450 nm) for the qualitative measurement of human Cytochrome P450 3A7 (CYP3A7) protein expression in cultured cells, with adherent cells as the sample type. The kit is described as lysate-free and high-throughput, and can be used to compare relative Cytochrome P450 3A7 levels between samples as well as to screen the effects of treatments, inhibitors (such as siRNA or chemicals) and activators on the target.

The assay uses an indirect ELISA format: a CYP3A7-specific primary antibody captures the target, and an HRP-conjugated secondary antibody catalyses a colorimetric reaction upon substrate addition. A monoclonal anti-human GAPDH antibody is included as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining step is used to determine cell density and adjust for plating differences. The stated dynamic range is > 5000 cells. The kit is configured for a 96-Well Microplate format and is recommended for storage at 4 °C with a stability of 6 months. The product is classified under the Metabolism research area, with reactivity confirmed for human samples.

Cytochrome P450 3A7 is a heme-thiolate monooxygenase involved in an NADPH-dependent electron transport pathway in liver microsomes, where it oxidises steroids, fatty acids and xenobiotics. The human gene (CYP3A7) is located on chromosome 7q22.1 and encodes a protein of 61,547 Da. Database references for the target include Gene ID 1551, UniProt ID P24462 and OMIM 605340.
