Description

Assay Genie offers a Cell-Based ELISA Kit designed for the detection and relative quantification of Cytochrome P450 39A1 (CYP39A1) in cultured cells. Using a lysate-free, indirect ELISA format, the assay captures target protein via specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to drive a colorimetric reaction.

The kit is reactive against human and mouse samples and is intended for use with adherent cell cultures. It can be applied to measure relative levels of CYP39A1 in cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), and activators on CYP39A1 expression. The colorimetric readout is measured at 450 nm, with a dynamic range of more than 5000 cells. For normalisation, a monoclonal anti-GAPDH antibody is provided as an internal positive control, and Crystal Violet whole-cell staining may be used to determine cell density.

CYP39A1 is a member of the cytochrome P450 superfamily of monooxygenases involved in drug metabolism and the synthesis of cholesterol, steroids and other lipids. The endoplasmic reticulum protein acts as 24-hydroxycholesterol 7-alpha-hydroxylase and plays a role in bile acid metabolism by contributing to the conversion of cholesterol to bile acids, with substrates including the oxysterols 24-, 25-, and 27-hydroxycholesterol. The kit is supplied in a microplate format and should be stored at 4 °C with a stated stability of 6 months.
