Description

Assay Genie's Cytochrome P450 2U1 Colorimetric Cell-Based ELISA is a lysate-free, high-throughput assay kit designed to detect Cytochrome P450 2U1 protein expression in cultured cells. The kit uses an indirect ELISA format in which Cytochrome P450 2U1 is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyse a colorimetric reaction upon substrate addition.

The kit is intended for measuring relative levels of Cytochrome P450 2U1 in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators. Because the assay is qualitative, two normalisation strategies are included: a monoclonal antibody specific for human GAPDH as an internal positive control, and a Crystal Violet whole-cell staining step to determine cell density and adjust for plating differences.

Reactivity is reported for human, mouse, and rat samples, with a dynamic range of > 5000 cells, colorimetric detection at 450 nm, and a 96-well microplate format suited to adherent cells. The kit is stored at 4°C with a stated stability of 6 months. The target enzyme, CYP2U1, is a member of the cytochrome P450 superfamily of monooxygenases that catalyses the hydroxylation of arachidonic acid, docosahexaenoic acid, and other long-chain fatty acids, and has been linked to Spastic Paraplegia 56, autosomal recessive, supporting research in drug metabolism, toxicology, and related fields.
