Description

A cell-based, colorimetric ELISA from Assay Genie designed for the qualitative detection of Cytochrome P450 2D6 protein in cultured human cells. The kit uses a lysate-free, indirect ELISA format in which the target is captured by Cytochrome P450 2D6-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction read at 450 nm. It is intended for measuring relative Cytochrome P450 2D6 expression in adherent cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), and activators. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining step can be used to adjust absorbance values to cell density. The reported dynamic range is > 5000 Cells, and the kit is stored at 4°C for 6 months.

Cytochrome P450 2D6 (gene symbol CYP2D6) is a member of the cytochrome P450 superfamily of monooxygenases that localizes to the endoplasmic reticulum and participates in the metabolism of many drugs, with reported substrates including antidepressants, antipsychotics, analgesics, antitussives, beta-adrenergic blocking agents, antiarrhythmics, and antiemetics. According to the NCBI summary text cited in the product datasheet, the enzyme is known to metabolize as many as 25% of commonly prescribed drugs. The kit supports research in the metabolism area, with applications in studies of drug metabolism, drug interactions, and personalized medicine, where variation in CYP2D6 activity can influence drug efficacy and safety.
