Description

This cell-based ELISA kit from Assay Genie provides a sensitive, high-throughput, lysate-free workflow for measuring Cytochrome P450 26A1 levels in cell lysates and tissue samples. It can be used to determine the relative expression profile of Cytochrome P450 26A1 in cultured cells and to screen the effects of treatments, inhibitors such as siRNA or chemicals, and activators on the target.

Detection follows an indirect ELISA format: target-specific primary antibodies capture Cytochrome P450 26A1, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction read at 450 nm. An included anti-GAPDH monoclonal antibody serves as an internal positive control for normalising target absorbance, while a Crystal Violet whole-cell staining step allows cell density to be measured so that plating differences can be adjusted.

The kit is reactive with human, mouse and rat samples and is suitable for adherent cells. It uses a 96-well microplate format with a dynamic range of > 5000 cells and is stored at 4 °C, where it remains stable for 6 months. Cytochrome P450 26A1, encoded by the gene CYP26A1 (UniProt O43174), is an endoplasmic reticulum-localised member of the cytochrome P450 superfamily that plays a key role in retinoic acid metabolism through 4-hydroxylation and 18-hydroxylation of retinoids including all-trans-retinoic acid and 9-cis-RA. Dysregulation of the enzyme has been linked to several diseases, making it a relevant target for pharmacology and toxicology research.
