Description

From Assay Genie, this colorimetric in-cell ELISA is designed for adherent cells and enables detection of cytochrome c protein expression in cultured cells. The assay uses an indirect format in which cytochrome c is captured by target-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition.

It is a lysate-free, high-throughput and sensitive method for measuring relative amounts of cytochrome c in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density after the colorimetric measurement.

The kit shows reactivity with Human, Mouse and Rat samples, with a dynamic range of > 5000 Cells. Cytochrome C is a mitochondrial protein central to the electron transport chain that plays a key role in apoptosis; its dysregulation has been linked to cancer, Alzheimer's and Parkinson's disease. The target corresponds to gene symbol CYCS (Gene ID: 54205; UniProt ID: P99999) with a molecular weight of 11,749 Da. Detection is by colorimetric measurement at 450 nm, with recommended storage at 4°C for 6 months.
