Description

A cell-based ELISA kit from Assay Genie enables researchers to profile Cyclin E2 protein expression directly in cultured cells. Designed for cell cycle research, the kit provides a lysate-free, high-throughput workflow for measuring the relative amounts of Cyclin E2 in cultured cells and for screening the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on Cyclin E2 expression.

The assay is configured in a 96-Well Microplate format with colorimetric detection at 450 nm and is reactive with Human and Mouse samples. It has a stated dynamic range of > 5000 Cells, a sample type of Adherent Cells, and components are stored at 4 °C with a stability of 6 Months. Detection follows an indirect ELISA format in which Cyclin E2 is captured by Cyclin E2-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody; the conjugated HRP then catalyses a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining method is used to determine cell density, allowing absorbance values to be normalised to cell amount to adjust for plating differences. The kit includes anti-phospho and anti-target antibodies, HRP-conjugated anti-rabbit IgG and anti-mouse IgG secondary antibodies, blocking and wash buffers, ready-to-use substrate, stop solution, and Crystal Violet solution, with absorbance read at 450 nm after stop solution addition.

The target, Cyclin E2, is encoded by the gene CCNE2 (Gene ID: 9134, UniProt ID: O96020), a member of the cyclin family and Cyclin E subfamily classified as a cell cycle regulator and activator. According to the supplied UniProt information, CCNE2 is essential for the control of the cell cycle at the late G1 and early S phase and forms a serine/threonine kinase holoenzyme complex with CDK2 and CDK3, with the cyclin subunit imparting substrate specificity to the complex.
