Description

Assay Genie offers a Cell-Based ELISA Kit designed for the measurement of Cyclin D1 phosphorylated at Thr286 in adherent cells. The kit targets Cyclin D1 (Phospho-Thr286) and is reactive with Human, Mouse, and Rat samples, supporting cell cycle research across multiple model systems.

Detection is performed by colorimetric measurement at 450 nm using an indirect ELISA format, with a dynamic range of > 5000 Cells. The phospho-specific assay is lysate-free and high-throughput, allowing detection of Cyclin D1 protein phosphorylation and expression profiles directly in cultured cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing absorbance readings, while Crystal Violet whole-cell staining can be used to determine cell density. HRP-conjugated secondary antibodies against rabbit and mouse IgG catalyze the colorimetric reaction upon substrate addition.

Cyclin D1 (gene symbol CCND1, Gene ID 595, UniProt ID P24385) is a member of the cyclin family and functions as a regulatory subunit of CDK4 and CDK6, whose activity is required for the G1/S transition of the mitotic cell cycle. The protein has a molecular weight of 34kDa and is classified as an oncoprotein involved in cell cycle regulation. Phosphorylation at Thr286 plays a crucial role in the stability and function of Cyclin D1, making it an important marker for studying cell proliferation and cancer development.
