Description

This Cell-Based ELISA Kit from Assay Genie is designed for the qualitative measurement of Cyclin D1 protein expression in cultured adherent cells. The assay uses an indirect ELISA format, in which Cyclin D1 is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm upon substrate addition. It is a convenient, lysate-free, high-throughput and sensitive method for profiling Cyclin D1 in cells, and can be applied to screen the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on Cyclin D1 levels.

Reactive with Human, Mouse and Rat samples, the kit has a stated dynamic range of "> 5000 Cells" and is stored at 4°C for 6 Months. Normalization is supported by two approaches: a monoclonal antibody specific for human GAPDH is included as an internal positive control, and a Crystal Violet whole-cell staining method can be used to determine cell density after the colorimetric HRP measurement, allowing absorbance values to be normalized to cell amounts.

Cyclin D1 (gene symbol CCND1, Gene ID: 595, UniProt ID: P24385, molecular weight 34kDa) is a member of the highly conserved cyclin family, functioning as a regulatory subunit of CDK4 or CDK6 and controlling the G1/S transition of the cell cycle. Mutations, amplification and overexpression of CCND1 are observed frequently in a variety of tumors and may contribute to tumorigenesis, making it a key biomarker for research into cell proliferation, cancer biology and related therapeutic interventions.
