Description

From Assay Genie, the Cyclin B1 (Phospho-Ser147) Colorimetric Cell-Based ELISA Kit is a cell-based, lysate-free assay for detecting phosphorylation of Cyclin B1 at serine 147 in cultured cells, with a colorimetric readout measured at 450 nm. It is supplied in a 96-well microplate format and is reactive with Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells. The intended sample type is adherent cells.

The kit uses an indirect ELISA format. Phospho-specific and total Cyclin B1 primary antibodies capture the target, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition, allowing qualitative determination of phosphorylated Cyclin B1. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences. The kit can be used to measure relative levels of phosphorylated Cyclin B1 in cultured cells and to screen the effects of various treatments, inhibitors (e.g. siRNA or chemicals), or activators on Cyclin B1 phosphorylation.

Cyclin B1 is a pivotal regulator of cell cycle progression, particularly the G2/M transition, and phosphorylation at serine 147 modulates its activity, influencing cell division and proliferation. Dysregulation of this process has been implicated in diseases such as cancer. The target corresponds to gene CCNB1 (Gene ID: 891, UniProt ID: P14635, OMIM: 123836, Unigene: Hs.23960), with a molecular weight of 44,102 Da and a chromosomal location of 5q12. CCNB1 complexes with cdc2 to form the maturation-promoting factor (MPF), and is classified as a cell cycle regulator and activator.
