Description

This cell-based ELISA kit from Assay Genie is designed for the detection of phosphorylated Cyclin B1 at Ser126 in cultured cells, offering high sensitivity and specificity for accurate, reproducible results across a variety of research applications. The kit enables measurement of the relative amounts of phosphorylated Cyclin B1 in cultured cells and can be used to screen the effects of various treatments, inhibitors (including siRNA or chemicals), and activators on Cyclin B1 phosphorylation.

The assay employs an indirect ELISA format in which Cyclin B1 (Phospho-Ser126) is captured by phospho-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyse a colorimetric reaction upon substrate addition. Detection is colorimetric, read at 450 nm. The kit is reactive with human, mouse and rat samples, with a dynamic range of > 5000 cells, and is intended for adherent cells. The procedure is lysate-free, providing a high-throughput approach to cellular analysis.

Because the cell-based ELISA is qualitative in nature, two normalisation methods are included: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density so that absorbance values can be normalised to cell amount, adjusting for plating differences. The target gene is CCNB1, classified as a Phospho sub-type, and the kit is intended for the Cell Cycle research area.
