CrkII (Phospho-Tyr221)Fluorometric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based Phospho ELISA Kit enables researchers to monitor CrkII (Phospho-Tyr221) protein phosphorylation and expression profiles directly in cells. The kit provides a lysate-free, high-throughput, and sensitive workflow for measuring the relative amounts of phosphorylated CrkII in cultured cell lines and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on CrkII phosphorylation.

Detection is achieved through an indirect ELISA format in which CrkII (Phospho-Tyr221) is captured by target-specific primary antibodies, with Dye 1-conjugated and Dye 2-conjugated secondary antibodies binding the Fc region of the primary antibody to enable a fluorometric readout at Ex/Em 651/667 nm and 495/521 nm. The assay is reactive with Human, Mouse, and Rat samples, with a stated dynamic range of greater than 5000 cells. Two normalization strategies are provided: a monoclonal antibody specific for human GAPDH serves as an internal positive control, and an antibody against the non-phosphorylated counterpart of CrkII is included to normalize RFU values.

The kit is supplied in a 96-well microplate format suited to cell-based ELISA workflows, with recommended storage at 4°C and a stated shelf life of 6 months.
