Description

A cell-based ELISA from Assay Genie for detecting CREM (cAMP-responsive element modulator) protein in cultured cells. The kit uses an indirect ELISA format with HRP-conjugated secondary antibodies and a colorimetric readout at 450 nm, providing a lysate-free, high-throughput and sensitive assay with high specificity for measuring relative CREM expression levels.

Reactivity is reported for human, mouse, and rat samples, with a stated dynamic range of > 5000 cells, and the protocol is intended for adherent cell cultures. A monoclonal anti-GAPDH antibody is included as an internal positive control to normalize target absorbance, and a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences. The qualitative assay is suited to studying CREM activation in response to treatments, inhibitors such as siRNA or chemicals, and activators.

CREM is a bZIP transcription factor that binds the cAMP responsive element in many viral and cellular promoters and is involved in cAMP-mediated signal transduction, with roles in spermatogenesis, circadian regulation of gene expression, and cell differentiation. Aberrant CREM activity has been implicated in cancer, immune disorders, and neurological conditions. The kit is supplied in a microplate format for cell biology research and is stored at 4 °C with a stated stability of 6 months.
