The CREB Transcription Factor Activity Assay from Assay Genie is a DNA-binding ELISA kit that quantifies the DNA-binding competence of activated CREB (cAMP response element-binding protein) in nuclear and whole-cell lysates. The 96 Assays microplate is pre-coated with a double-stranded CRE consensus oligonucleotide (5'-TGACGTCA-3'), so only CREB that is properly dimerized and capable of engaging its response element is captured and detected. Bound transcription factor is revealed with a CREB-specific antibody and a chromogenic substrate, providing a quantitative readout of pathway activation rather than total protein level.

This makes the kit useful for probing cAMP/PKA, calcium, MAPK, and growth-factor signaling in cell-culture and tissue models, including studies of neuronal plasticity, learning and memory, glucose homeostasis, hormone responses, and oncogenic transformation. The assay is validated for human, mouse, and rat samples and accepts both nuclear extracts and whole-cell lysates, so it can be paired with upstream manipulations such as forskolin, IBMX, BDNF, or kinase inhibitors. Typical workflows use roughly 5 to 20 ug of nuclear protein per well, with results normalized to a wild-type CRE oligonucleotide competitor to control for binding specificity.

Intended for research use only and in vitro applications. Store reagents at -20 degrees C, protect the substrate from light, and avoid repeated freeze-thaw cycles of the antibody and oligonucleotide components. Each kit provides a full 96 Assays plate, sufficient for two independent 8-point curves or 12 to 24 sample conditions in duplicate.
