Creatinine is a nitrogenous waste product generated from the spontaneous, non-enzymatic cyclization of creatine and creatine phosphate in muscle tissue, and its circulating concentration is one of the most widely used biochemical indicators of glomerular filtration rate, renal function, and whole-body protein turnover. This Assay Genie competitive immunoassay enables quantitative determination of creatinine using a microtitre plate format, in which analyte present in the specimen competes with a conjugated creatinine derivative for binding to anti-creatinine antibody immobilized on the plate surface.

The kit supplies pre-coated wells, a set of calibrators, enzyme conjugate, concentrated wash buffer, chromogenic substrate, and stop solution. A 50 µL aliquot of serum or plasma is loaded per well alongside the provided standards, and the complete assay is typically finished within approximately 90 minutes at room temperature. After incubation and washing steps, the substrate is added and absorbance is read on a standard microplate reader.

A freshly prepared standard curve is recommended for each plate, and any sample exceeding the highest calibrator should be diluted in assay buffer and re-assayed. Unopened kit components should be stored refrigerated as indicated in the product insert; reconstituted reagents remain stable for the period specified in the manual.
