Assay Genie's COVID-19 Human IgM PharmaGenie ELISA Kit is built around recombinant SARS-CoV-2 nucleocapsid (N) protein, a highly immunogenic structural antigen expressed abundantly during viral replication. By coating the microplate with this N-protein antigen, the Indirect ELISA captures circulating anti-N IgM antibodies in patient serum, enabling qualitative detection of recent or acute infection.

IgM-class antibodies typically emerge within 1-3 weeks after symptom onset, making this assay particularly informative for early-stage serological screening and complementary to molecular PCR diagnostics. The Indirect ELISA workflow relies on an enzyme-conjugated anti-human IgM secondary antibody, uses a 10 µL serum sample per well, and produces absorbance readings proportional to anti-N IgM titers in the specimen.

Because the assay targets the N-protein rather than the spike (S) protein, it provides a distinct immunological readout: anti-N IgM responses frequently arise earlier and can persist longer in many patients, supporting seroprevalence studies, vaccine response profiling, and surveillance work in research settings. The kit is intended for in vitro research use only and is configured as a microplate-format assay with a colorimetric readout compatible with routine laboratory instrumentation.
