Description

Manufactured by STEMCELL Technologies, this lyophilized dissociation reagent is a protease preparation derived from Clostridium histolyticum. The enzyme consists of a single polypeptide chain of approximately 1000 amino acid residues in length and is capable of digesting native collagen fibrils commonly found in connective tissues, making it frequently used for tissue dissociation. The preparation contains the combined activity of several proteases, including collagenase, caseinase, clostripain, and trypsin. With its low tryptic activity, it is well suited to applications such as the dissociation of pancreatic islets and to workflows where maintenance of membrane receptor integrity is required.

Supplied as a lyophilized powder, the reagent is stored at 2 - 8 °C and remains stable as supplied for 12 months from date of receipt; it can be reconstituted in a balanced salt solution or buffer of choice. Reported molecular weight is 68 - 130 kDa, with an optimum pH of 6.3 - 8.5 and cleavage at the -Pro-X-✝-Gly-Pro-Y- sequence, where X is a neutral residue and Y is nonspecific. Activity specifications state collagenase at ≥ 160 CDU/mg dry weight, caseinase at ≥ 100 u/mgdw, clostripain at ≤ 3.0 u/mgdw, and trypsin at ≤ 0.1 u/mgdw.
