Crude collagenase enzyme preparation derived from Clostridium histolyticum, formulated for gentle dissociation of mammalian tissue in primary cell isolation workflows. The blend retains collagenase as the principal activity alongside caseinase, clostripain, and a trypsin-like protease, giving a balanced digestion profile useful for soft, connective, and glandular tissues including rodent and human pancreas islet isolation as well as adult hepatocyte preparation. It is commonly paired with DNase I and a low-calcium wash to reduce clumping in single-cell suspensions, then filtered through a 70 µm cell strainer before plating or downstream analysis.
Supplied as a 100 mL bottle of lyophilized powder that is reconstituted in HBSS or DMEM to a working concentration of 0.5 to 2.5 mg/mL depending on tissue density. Digests are typically carried out at 37 °C for 15 to 45 minutes with gentle rocking, followed by quenching with cold buffer. Store the dry powder at -20 °C, protected from light, and keep reconstituted aliquots frozen at -20 °C for up to one month to limit freeze-thaw loss of activity. This reagent from STEMCELL Technologies is intended for research use only.
