Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the accurate detection of Collagen VI alpha2 levels in cell culture supernatants. The assay uses a colorimetric detection method read at 450 nm and is reactive with both human and mouse samples, with a stated dynamic range of > 5000 Cells.

The kit employs an indirect ELISA format that is lysate-free, high throughput, and sensitive for detecting Collagen VI alpha2 protein expression profiles in cells. It can be used to measure relative amounts of Collagen VI alpha2 in cultured cells and to screen for the effects of various treatments, inhibitors such as siRNA or chemicals, or activators. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values. Following colorimetric measurement of HRP activity via substrate addition, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts and plating differences to be adjusted.

Collagen VI alpha2 is a key component of the extracellular matrix, with roles in tissue development and maintenance. Dysregulation has been linked to diseases including Bethlem myopathy, Ullrich congenital muscular dystrophy, and autosomal recessive myosclerosis, making it a valuable biomarker for studying these pathologies. The kit is supplied in a 96-well microplate format, stored at 4°C, and stable for 6 months.
