Collagen IV Colorimetric Cell-Based ELISA Kit

This cell-based ELISA kit from Assay Genie is designed for the qualitative detection and relative quantification of Collagen IV protein expression in cultured cells, using a lysate-free, high-throughput workflow. The assay uses an indirect ELISA format in which Collagen IV is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition; absorbance is read at 450 nm.

A monoclonal anti-GAPDH antibody is included as an internal positive control, and a Crystal Violet whole-cell staining step is provided to normalise absorbance to cell density, allowing comparison across wells with different plating or treatment conditions. Collagen IV is encoded by the COL4A1 gene (Gene ID: 1282; UniProt ID: P02462; Molecular Weight: 127,981 Da) and is the major structural component of glomerular basement membranes, forming a 'chicken-wire' meshwork together with laminins, proteoglycans and entactin/nidogen. Abnormalities in collagen IV expression have been linked to kidney disorders, muscular dystrophies, and certain types of cancer.
