Description

From Assay Genie, this cell-based ELISA kit is designed for the detection of cofilin protein in cultured cells using a colorimetric readout at 450 nm. The kit is configured in a 96-well microplate format and is reactive with human, mouse, and rat samples, with procedures optimised for adherent cells. A dynamic range of > 5000 cells is specified by the manufacturer, and the kit is stored at 4 °C with a stated stability of 6 months.

The assay uses an indirect ELISA format in which cofilin is captured by target-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyse a colour change upon substrate addition. Because the assay is qualitative, two normalisation methods are provided: a monoclonal anti-GAPDH antibody serves as an internal positive control, and a Crystal Violet whole-cell staining step allows absorbance values to be normalised to cell density, correcting for plating differences. The kit can be used to measure relative cofilin levels in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators.

Cofilin is a key regulatory protein involved in actin cytoskeleton dynamics, cell migration, and cell shape changes. Dysregulation of cofilin has been associated with diseases such as cancer, Alzheimer's, and heart disease, making it a biomarker of interest in studies of cellular processes and disease pathogenesis. The target corresponds to gene symbols CFL1/CFL2 (Gene IDs 1073/1072; UniProt P23528).
