Description

This Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the precise measurement of phosphorylated Claudin-6 at tyrosine 219 in cell lysates. It provides a lysate-free, high-throughput workflow for detecting Claudin-6 protein phosphorylation and expression profiles directly in cultured cells, and is suitable for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Claudin-6 phosphorylation.

Qualitative detection is achieved through an indirect ELISA format, in which Claudin 6 (Phospho-Tyr219) is captured by target-specific primary antibodies and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. The assay is read at 450 nm using a colorimetric detection method. Reactivity has been validated for Human and Mouse samples, and the dynamic range is greater than 5,000 cells. A monoclonal anti-human GAPDH antibody is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining step can be used to adjust absorbance values to cell density.

Claudin-6 is a tight-junction membrane protein that plays a major role in cell-cell adhesion, barrier function, and signal transduction. The gene symbol is CLDN6 (Gene ID: 9074, UniProt ID: P56747), with the human ortholog located at chromosomal position 16p13.3. Because phosphorylation at tyrosine 219 is known to regulate Claudin-6 function and has been implicated in disease processes, quantifying phosphorylated Claudin-6 offers insight into signalling pathways relevant to cancer, cardiovascular disease, and neurodegenerative disorders. The kit is intended for the Immunology research area and is used with adherent cells.
