Claudin 5 Colorimetric Cell-Based ELISA Kit

An Assay Genie cell-based ELISA kit enables qualitative, lysate-free measurement of Claudin 5 protein expression directly in cultured cells. The assay is designed for adherent cells and is suitable for quantifying relative Claudin 5 levels as well as screening the effects of treatments, inhibitors such as siRNA or chemicals, and activators on Claudin 5 expression.

Detection follows an indirect ELISA format: Claudin 5 is captured by target-specific primary antibodies and bound by HRP-conjugated secondary antibodies, producing a colorimetric reaction read at 450 nm. A monoclonal GAPDH antibody is supplied as an internal positive control for normalization, and a Crystal Violet whole-cell staining step is used to determine cell density. The kit is reactive with human, mouse, and rat samples, has a dynamic range of > 5000 Cells, and is supplied in a 96-Well Microplate format for storage at 4°C for 6 months.

Claudin-5 is a tight-junction protein that supports blood-brain barrier function and vascular integrity in endothelial cells, and its dysregulation has been associated with neuroinflammation, neurological disorders, and cancer, making it a relevant target in research and therapeutic development.
