Claudin 3 Colorimetric Cell-Based ELISA Kit

An in-cell ELISA kit from Assay Genie designed to detect and quantify levels of Claudin-3 in cultured cell samples. The assay uses a colorimetric detection method read at 450 nm in a 96-well microplate format and is described as lysate-free, high-throughput and sensitive for measuring relative Claudin-3 expression in adherent cells. Qualitative determination is achieved by an indirect ELISA format, in which Claudin-3 is captured by specific primary antibodies and HRP-conjugated secondary antibodies catalyse a colour reaction upon substrate addition.

Reactivity is reported for human, mouse and rat, with a dynamic range of > 5000 cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and following the colorimetric read, Crystal Violet whole-cell staining may be used to determine cell density so absorbance values can be normalised to cell amount. The kit is stored at 4 °C and is reported stable for 6 months under those conditions.

Claudin-3 is an integral membrane protein of the claudin family and a component of tight junction strands, playing a major role in calcium-independent cell-cell adhesion. Dysregulation of Claudin-3 has been linked to diseases including cancer and inflammatory bowel disease, making the kit a relevant tool for researchers studying cell junctions, barrier function and cellular pathophysiology.
