Description

From Assay Genie, the Claudin 11 Colorimetric Cell-Based ELISA Kit is a convenient, lysate-free, high-throughput and sensitive cell-based ELISA designed to detect the protein expression profile of Claudin 11 in cells. It can be used to measure the relative amounts of Claudin 11 in cultured cells and to screen the effects that various treatments, inhibitors (such as siRNA or chemicals) or activators have on Claudin 11.

Qualitative determination of Claudin 11 concentration is achieved by an indirect ELISA format: Claudin 11 is captured by Claudin 11-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The conjugated HRP enzyme catalyses a colorimetric reaction upon substrate addition. The kit is reactive with Human, Mouse and Rat samples, has a stated dynamic range of > 5000 Cells, and uses colorimetric detection at 450 nm. It is supplied in a microplate format, intended for storage at 4°C with a stability of 6 Months. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density so that plating differences can be adjusted.

Claudin-11 is a key protein involved in cell-to-cell adhesion and the formation of tight junctions, regulating the permeability of epithelial and endothelial barriers. Dysfunction of Claudin-11 has been implicated in autoimmune disorders, inflammatory conditions and cancer, making it a critical target for research and therapeutic development. According to the cited UniProt entry (UniProt ID: O75508; Gene ID: 5010; molecular weight 22kDa), the protein plays a major role in tight junction-specific obliteration of the intercellular space through calcium-independent cell-adhesion activity and belongs to the claudin family.
