Claudin 10 Colorimetric Cell-Based ELISA Kit

Assay Genie offers a Cell-Based ELISA Kit for the qualitative measurement of Claudin 10 in cell culture samples. Designed for use with adherent cells, the kit employs a colorimetric detection method read at 450 nm and is reactive with human and mouse samples, with a stated dynamic range of > 5000 cells. It provides a lysate-free, high throughput workflow for profiling Claudin 10 protein expression in cultured cells and for screening the effects of various treatments, inhibitors, or activators on the target.

The assay is built on an indirect ELISA format in which Claudin 10 is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of those primaries. The bound HRP then catalyses a colour-forming reaction upon substrate addition. Two normalisation strategies are built in: a monoclonal anti-human GAPDH antibody serves as an internal positive control, and Crystal Violet whole-cell staining can be used to account for cell density after the colorimetric read.

Claudin-10 is a tight-junction protein that helps maintain the integrity of epithelial cell barriers, and its dysregulation has been linked to inflammatory disorders and cancer. The gene, CLDN10, maps to chromosomal region 13q32.1 and encodes a 24 kDa member of the claudin family. Storage is at 4°C with a stated stability of 6 months, and the kit is intended for research applications in signal transduction.
