Description
From Assay Genie, this cell-based ELISA kit is designed for the qualitative measurement of Claudin-1 protein expression directly in cultured adherent cells, without the need to prepare lysates. It uses an indirect ELISA format in which Claudin-1 is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies, producing a colorimetric signal read at 450 nm. The kit is suitable for profiling relative Claudin-1 levels and for screening the effects of various treatments, inhibitors such as siRNA or chemicals, and activators.
A monoclonal anti-GAPDH antibody is included as an internal positive control to normalize target absorbance, and Crystal Violet whole-cell staining may be used to determine cell density, allowing absorbance values to be normalized to cell amounts. The stated dynamic range is greater than 5,000 cells, with reactivity demonstrated in human, mouse and rat samples. Storage stability is given as 4 °C for 6 months. Claudin-1 is an integral membrane protein and component of tight junction strands that mediates calcium-independent cell-cell adhesion and acts as a co-receptor for HCV entry into hepatic cells; it is strongly expressed in liver and kidney.
Database identifiers associated with the target include Gene ID 9076, UniProt ID O95832, OMIM entries 603718 and 607626, and Unigene Hs.439060, with the human gene located at chromosomal region 3q28-q29.
