CKMT2 Colorimetric Cell-Based ELISA

A colorimetric cell-based ELISA kit from Assay Genie is designed to detect and quantify CKMT2 protein levels in cell lysates and tissue extracts. The kit uses an indirect ELISA format: CKMT2-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, enabling a colorimetric reaction upon substrate addition. It is described as a lysate-free, high-throughput and sensitive assay that can detect CKMT2 protein expression profiles in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on CKMT2.

Detection is measured at 450 nm, with a dynamic range of > 5000 Cells. The kit is reactive with Human, Mouse and Rat samples and is suited to adherent cells. Storage is at 4°C for 6 months. A monoclonal anti-human GAPDH antibody is included as an internal positive control to normalize target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density, by which plating differences can be adjusted.

CKMT2, also known as mitochondrial creatine kinase, is a key enzyme involved in energy metabolism within mitochondria, playing a crucial role in the generation of ATP. It belongs to the ATP:guanido phosphotransferase family and reversibly catalyzes the transfer of phosphate between ATP and various phosphogens such as creatine phosphate. Creatine kinase isoenzymes play a central role in energy transduction in tissues with large, fluctuating energy demands, including skeletal muscle, heart, brain and spermatozoa. The gene maps to chromosomal location 5q13.3 and encodes a mitochondrial protein of 47,504 Da (UniProt ID P17540, Gene ID 1160), making this kit ideal for research into mitochondrial dysfunction, muscle diseases, and the role of CKMT2 in various cellular processes.
