Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the accurate detection of CKII beta phosphorylation at Ser209 in cell lysates. The colorimetric assay is read at 450 nm and is reactive with human, mouse, and rat samples, with a stated dynamic range of > 5000 Cells. It is intended for use with adherent cell cultures.

The kit uses a lysate-free, indirect ELISA format in which CKII-beta (Phospho-Ser209) is captured by phospho-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density for further normalization. Researchers can use the kit to measure relative levels of phosphorylated CKII-beta in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on CKII-beta phosphorylation.

The target, CKII-beta (gene symbol CSNK2B), is the regulatory beta subunit of the casein kinase II holoenzyme, which participates in Wnt signaling and influences cellular processes such as cell growth, survival, and differentiation. Dysregulation of this phosphorylation event has been implicated in cancer, neurodegenerative disorders, and inflammatory conditions, making it a target of interest in metabolism-related research. The kit is classified as a phospho-specific Cell-Based ELISA.
