Description

From Assay Genie, this cell-based ELISA kit is designed for qualitative measurement of CKII-beta protein expression in cultured cells. It uses an indirect ELISA format in which CKII-beta is captured by target-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm upon substrate addition. The procedure is lysate-free and high-throughput, suitable for measuring relative CKII-beta amounts in cultured cells and for screening the effects of treatments, inhibitors such as siRNA or chemicals, or activators on CKII-beta expression.

The kit is reactive with human, mouse and rat samples and is recommended for use with adherent cells, supporting a dynamic range of > 5000 Cells. It is supplied in a 96-well microplate format with a colorimetric readout. For data normalization, a monoclonal anti-GAPDH antibody is included as an internal positive control, and a Crystal Violet whole-cell staining step can be used to determine cell density so that absorbance values can be adjusted for plating differences.

CKII Beta, also known as casein kinase II, is involved in regulating cell growth, DNA replication and cell differentiation, and its dysregulation has been linked to cancer, neurodegenerative disorders and cardiovascular diseases. The assay targets the protein encoded by CSNK2B (Gene ID 1460; UniProt P67870), the regulatory beta subunit of the casein kinase 2 holoenzyme, with a reported molecular weight of 24,942 Da. The kit is intended for metabolism research and is stored at 4 °C with a stated stability of 6 months.
