Description

A cell-based ELISA kit from Assay Genie provides a lysate-free, high-throughput and sensitive method for detecting the CKI-epsilon protein expression profile in cells. The assay uses an indirect ELISA format: CKI-epsilon is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyse a colorimetric reaction upon substrate addition, with absorbance read at 450 nm.

The kit enables measurement of the relative amounts of CKI-epsilon in cultured adherent cells and screening for the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on the target. An anti-GAPDH monoclonal antibody is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining is provided to determine cell density for additional normalisation.

CKI-epsilon (casein kinase I isoform epsilon, encoded by CSNK1E) is a ubiquitous serine/threonine protein kinase of the CK1 family. It is a central component of the circadian clock, and together with Dvl-1 and Frat-1 it activates the Wnt signalling pathway. The kinase may act as a negative regulator of circadian rhythmicity by phosphorylating PER1 and PER2 and is implicated in cell cycle progression. Reported reactivity is for human and mouse samples, with a dynamic range of greater than 5,000 cells, and the kit is intended for the cell biology research area. Storage is at 4 °C for up to six months.
