Description

Manufactured by Assay Genie, this cell-based ELISA kit is designed to detect and measure Chk2 protein phosphorylated at threonine 68 directly in cultured cells, without the need for lysate preparation. The assay uses an indirect ELISA format in which Chk2 (Phospho-Thr68) is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyse a colorimetric reaction, read at 450 nm.

The kit has been validated for human, mouse and rat samples and accepts adherent cells, with a dynamic range of > 5000 Cells. It is suitable both for measuring relative levels of phosphorylated Chk2 in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals) and activators on Chk2 phosphorylation. Because the assay is qualitative, two normalization methods are provided: a monoclonal anti-GAPDH antibody serves as an internal positive control, and a Crystal Violet whole-cell staining step is used to determine cell density so that absorbance values can be adjusted for plating differences.

Chk2 is a serine/threonine kinase (gene symbol CHEK2, UniProt O96017) that functions as a key regulator of the DNA damage response and a putative tumour suppressor. Phosphorylation at Thr68 is a critical activating event that allows Chk2 to coordinate cell-cycle arrest, DNA repair and apoptosis following DNA damage, making this phospho-specific assay a relevant tool for studies of DNA damage signalling and cancer-related research.
