Chk2 (Phospho-Thr383)Colorimetric Cell-Based ELISA Kit

Assay Genie provides a cell-based ELISA kit designed to detect and quantify levels of phosphorylated CHK2 (Checkpoint Kinase 2) at Thr383 in cells. The kit uses a colorimetric detection method with measurement at 450 nm in an indirect ELISA format, where target-specific primary antibodies capture the analyte and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition.

The assay is lysate-free, high throughput, and sensitive, allowing researchers to measure relative amounts of phosphorylated Chk2 in cultured cells. It can also be used to screen the effects of various treatments, inhibitors such as siRNA or chemicals, or activators on Chk2 phosphorylation. The kit is reactive with Human, Mouse, and Rat samples, with a dynamic range of > 5000 cells.

Two normalization methods are provided: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalizing target absorbance values, while a Crystal Violet whole-cell staining method can be used to determine cell density and adjust for plating differences. The kit is intended for adherent cell samples.
