Description

A Colorimetric Cell-Based ELISA Kit (In-Cell ELISA) from Assay Genie is designed to detect Chk2 protein phosphorylated at serine 516 in adherent cell samples. The assay uses a lysate-free, indirect ELISA format in which phospho-Ser516-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. Reactivity is reported for human samples, with a dynamic range of > 5000 Cells and detection measured at 450 nm.

The kit supports qualitative measurement of Chk2 (Phospho-Ser516) levels in cultured cells and can be used to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on Chk2 phosphorylation. Because the Cell-Based ELISA is qualitative, two normalization approaches are included: a monoclonal anti-GAPDH antibody serves as an internal positive control for normalizing target absorbance, and Crystal Violet whole-cell staining is used to determine cell density so absorbance values can be adjusted for plating differences.

Chk2 (gene symbol CHEK2) is described as a CAMK protein kinase of the Rad53 family and a cell cycle checkpoint regulator. It is rapidly phosphorylated in response to DNA damage and replication blocks, and phosphorylation at serine 516 is a key event in its activation and signaling cascade, making it a target of interest for studies of cellular stress responses, DNA repair mechanisms, and cancer progression.
