Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is designed for detecting Chk2 protein expression profiles directly in cultured cells. Using a lysate-free, indirect ELISA format, the kit offers high sensitivity and specificity and supports high-throughput screening of adherent cell samples. Detection relies on HRP-conjugated secondary antibodies catalysing a colorimetric reaction measured at 450 nm.

Reactive with human, mouse, and rat samples, the assay has a dynamic range covering more than 5000 cells and is suited to measuring relative Chk2 levels in cultured cells as well as screening the effects of treatments, inhibitors such as siRNA or chemical compounds, and activators. A monoclonal anti-human GAPDH antibody is included as an internal positive control for normalisation of absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density. The kit is stored at 4 °C with a stability of 6 months.

Chk2 (gene symbol CHEK2) is a serine/threonine kinase of the CAMK/RAD53 family that functions as a cell cycle checkpoint regulator and putative tumour suppressor. It is rapidly phosphorylated in response to DNA damage and replication blocks and acts on substrates including CDC25C, p53, and BRCA1 to halt cell cycle progression and promote DNA repair. Loss-of-function mutations in Chk2 are associated with Li-Fraumeni syndrome as well as breast, prostate, and other cancers, making it a valuable biomarker for research into DNA damage response pathways and potential therapeutic interventions.
