Description

Assay Genie's Cell-Based ELISA Kit is designed for the precise detection of phosphorylated CHK1 at serine 280 in cell samples, offering qualitative determination through a colorimetric method read at 450 nm. The assay uses an indirect ELISA format in which Chk1 (Phospho-Ser280) is captured by specific primary antibodies, and an HRP-conjugated secondary antibody catalyses a colorimetric reaction upon substrate addition.

The kit can be used with adherent cells and has a stated dynamic range of > 5000 Cells, making it suitable for measuring the relative amounts of phosphorylated Chk1 in cultured cells as well as screening for the effects that various treatments, inhibitors such as siRNA or chemicals, or activators have on Chk1 phosphorylation. CHK1 is a key regulator of cell cycle checkpoints and the DNA damage response, and phosphorylation at serine 280 is a post-translational modification that can impact CHK1 function and signalling pathways relevant to cell cycle regulation and DNA repair. The kit is reactive with human samples (Gene ID: 1111; UniProt ID: O14757).

For normalisation, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and Crystal Violet whole-cell staining may be used to determine cell density so that absorbance values can be normalised to cell amounts.
