The Assay Genie Chicken S100-A10 (S100A10) ELISA Kit quantitatively measures S100A10 protein levels in chicken serum and plasma samples using a sandwich immunoassay format. S100-A10, also referred to as p11 or calpactin I light chain, is a member of the EF-hand calcium-binding protein family that typically forms a heterotetrameric complex with annexin A2, contributing to membrane-cytoskeleton interactions, receptor trafficking, and ion channel regulation at the cell surface.

Each 96 assays kit includes a pre-coated microplate, lyophilized reference standards, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer, and TMB substrate solution. The colorimetric endpoint is read at 450 nm with an optional 570 nm wavelength correction, supporting accurate interpolation against a standard curve generated in every run. The assay is validated for chicken serum and plasma; cell culture supernatants and tissue homogenates may also be compatible after proper validation.

For reliable results, store unopened kit components at 2-8 °C, keep the microplate sealed and protected from light, and avoid repeated freeze-thaw cycles of reconstituted reagents. Reconstituted standards and diluted samples should be aliquoted and frozen at -80 °C when not used immediately. Intended for research use only in avian immunology, inflammation, infectious disease, and developmental biology studies.

S100A10 is a small (~11 kDa) EF-hand calcium-binding protein that is constitutively active in calcium binding even in the resting intracellular environment, distinguishing it from most other S100 family members. It functions primarily through its obligate heterotetrameric association with annexin A2 (ANXA2), forming the (S100A10:ANXA2)2 complex that localizes to the plasma membrane and endosomal compartments. This complex recruits plasminogen and tissue plasminogen activator to the cell surface, supporting localized fibrinolysis, and regulates trafficking and surface stability of several ion channels and receptors, including TASK-1 (K2P3.1) potassium channels, TRPV5/TRPV6 calcium channels, the serotonin 5-HT1B receptor, and the ASIC1a proton-gated channel.

In avian models, S100A10 expression has been characterized in chicken immune tissues and is implicated in inflammatory cytokine signaling, macrophage activation, and responses to avian pathogens. The Assay Genie sandwich ELISA is calibrated against purified recombinant chicken S100A10 standard, and the biotin-streptavidin-HRP detection chemistry provides amplified signal over direct-conjugate assays. Typical intra-assay CV remains below 10% and inter-assay CV below 12% when the protocol is followed as written. Recommended sample volume is 50-100 ul per well, with a total assay runtime of approximately 3.5 hours excluding optional overnight incubation steps. The pre-coated 96-well microplate strip format allows running partial plates; unused strips should be resealed with the desiccant pack and stored at 2-8 °C for stability through the kit expiration.

Researchers studying avian reproductive biology should note that S100A10 is expressed in the oviduct and has been linked to eggshell mineralization pathways, while studies of chicken embryonic fibroblasts have used S100A10 as a marker of membrane-cytoskeleton remodeling during development.
