The kit uses a colorimetric detection method and is intended for research applications requiring accurate and reproducible NGF quantitation. According to the manufacturer, the kit has a sensitivity of 0.281 ng/mL and a measuring range of 0.469–30 ng/mL, and is stored at 2–8°C for 12 months. Spiked-sample recovery averages 98% in serum (range 89–104%), 93% in EDTA plasma (range 85–97%), and 96% in heparin plasma (range 89–104%). Linearity across serial dilutions of 1:2, 1:4 and 1:8 falls within 95–105%, 85–104% and 97–105% respectively for serum, 85–100%, 83–98% and 82–100% for EDTA plasma, and 87–91%, 80–94% and 82–99% for heparin plasma.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks on the pre-coated plate and incubating at 37°C for 90 minutes for antigen binding. Biotin-labeled detection antibody is then added for a 60-minute incubation at 37°C, followed by HRP-Streptavidin (SABC) for 30 minutes. TMB substrate is added and incubated in the dark for 10–20 minutes, then the stop solution is added and absorbance is read at 450 nm.
