The Chicken Myoglobin (MB) ELISA Kit from Assay Genie is a sandwich-format colorimetric immunoassay that quantifies myoglobin levels in chicken serum and plasma samples. Myoglobin is a heme-containing protein found in skeletal and cardiac muscle, and circulating concentrations can serve as a biomarker of muscle injury, exercise stress, or inflammatory processes in avian research models.

This 96-well microplate ELISA relies on a sandwich configuration, with capture antibodies pre-coated on the plate. Standards and samples are added, followed by a biotinylated detection antibody and a streptavidin-HRP conjugate. A chromogenic substrate is then introduced, producing a measurable color change proportional to myoglobin concentration that is quantified on a standard microplate reader.

For sample handling, serum and plasma should be collected by standard venipuncture methods, aliquoted to limit freeze-thaw cycles, and stored refrigerated at 2-8 °C for short-term use or frozen at -20 °C for longer-term preservation. This kit is for research use only and is not validated for clinical diagnostic procedures.

Assay Genie supplies each kit with a pre-coated 96-well microplate, lyophilized chicken myoglobin standards, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer concentrate, TMB substrate, and stop solution. The capture and detection antibody pair is calibrated against chicken skeletal muscle myoglobin (a ~17 kDa heme protein), supporting specific quantitation with minimal cross-reactivity to hemoglobin or other avian serum proteins under standard conditions.

For the assay workflow, 50–100 µL of standards and diluted samples are loaded in duplicate wells, followed by sequential 30–60 minute incubations at 37 °C with the biotinylated antibody and streptavidin-HRP. After TMB development and addition of the stop solution (typically 2 M sulfuric acid), absorbance is read at 450 nm, with the standard curve fitted using a four-parameter logistic model. The typical dynamic range spans 1.5–100 ng/mL, with intra-assay CV reported below 8% and inter-assay CV below 10% in chicken matrices.

This kit is commonly applied in poultry nutrition, heat-stress, and muscle-physiology research, where circulating myoglobin is monitored as an indicator of skeletal muscle damage, transport stress, or exercise load in broilers, layers, and other avian research lines. Unopened kits are refrigerated at 2-8 °C and remain stable through the printed expiration, while reconstituted reagents should be aliquoted and frozen at -20 °C to limit freeze-thaw degradation.
