Chicken IL-15 ELISA Kit

A colorimetric Sandwich ELISA from Assay Genie, designed for the quantitative measurement of interleukin-15 in chicken samples, uses a double-antibody detection format. The kit is reactive with chicken IL-15 and validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. It offers a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL, and is supplied on a solid-phase microplate format intended for storage at 2-8°C for 12 months.

The assay follows a six-step protocol: equilibrate reagents and the pre-coated plate, then load standards, samples and a zero control and incubate at 37°C for 90 minutes; add biotin-labeled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; add TMB substrate and incubate in the dark for 10-20 minutes; then add stop solution and read absorbance at 450 nm. Sample preparation guidance covers serum (clot, centrifuge at 1000 × g for 20 minutes), plasma (collect in anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2-8°C), tissue homogenate, cell culture supernatant (centrifuge at 2500 rpm for 5 minutes) and cell lysate.

Linearity in serum (n=5) was 89-100% at 1:2, 91-98% at 1:4 and 81-91% at 1:8; in EDTA plasma (n=5) 87-95% at 1:2, 85-100% at 1:4 and 87-96% at 1:8; and in heparin plasma (n=5) 85-95% at 1:2, 91-99% at 1:4 and 80-94% at 1:8. Recovery was 87-105% (average 93%) for serum, 87-99% (average 95%) for EDTA plasma, and 92-104% (average 99%) for heparin plasma (n=5 each). IL-15 is catalogued under Uniprot F1NZN5 and is recognised as a cytokine, also referred to by the synonym IL15.
