Caspase 8 (CASP8) is an initiator cysteine-aspartic protease that drives the extrinsic apoptotic pathway upon engagement of cell-surface death receptors, including Fas, TRAIL receptors, and TNF receptor 1. Activation at the death-inducing signaling complex leads to cleavage of downstream effector caspases and execution of programmed cell death. This Assay Genie chicken CASP8 ELISA kit enables quantitative measurement of CASP8 protein in avian samples, supporting research into apoptosis regulation in poultry immunology, viral pathogenesis, and developmental biology.

The kit is configured for 96 individual assay determinations using a matched antibody pair raised against the chicken caspase 8 protein, allowing reproducible quantification across experimental conditions such as Marek's disease virus infection, avian influenza challenge, Salmonella enterica infection models, and chemical-induced apoptosis in chick embryo or primary hepatocyte systems. Reagents are supplied in individual vials sufficient for a full microplate run in duplicate.

For sample preparation, collect whole blood using standard anticoagulant or clot-activation protocols and centrifuge to obtain clarified supernatant fractions; flash-freeze aliquots and store at -80 °C to preserve caspase integrity, avoiding repeated freeze-thaw cycles. Equilibrate all kit components to room temperature before beginning the assay procedure, and run standards in parallel with experimental samples on the same plate to ensure accurate interpolation of unknowns.

Beyond poultry infectious disease research, this chicken CASP8 reagent supports work on oxidative stress in avian hepatocytes and comparative immunology studies of programmed cell death across vertebrate species. Researchers investigating Fas ligand signaling, TRAIL-mediated cytotoxicity, or death receptor clustering in avian cell lines will find quantitative CASP8 measurement a useful complement to western blot and flow cytometry readouts.
