Description

Assay Genie provides a cell-based ELISA kit designed for the qualitative measurement of CDK7 protein phosphorylation at threonine 170 in cultured cells. The assay uses a colorimetric detection method measured at 450 nm in an indirect ELISA format, in which a CDK7 (Phospho-Thr170)-specific primary antibody is captured and detected with an HRP-conjugated secondary antibody; the bound HRP then catalyses a colorimetric reaction upon substrate addition.

The lysate-free, high-throughput format is suitable for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on CDK7 phosphorylation. It is reactive with human and mouse samples, with a stated dynamic range of > 5000 Cells. A monoclonal anti-GAPDH antibody is included as an internal positive control to normalize target absorbance values, and Crystal Violet whole-cell staining can be applied after the colorimetric readout to determine cell density and adjust for plating differences. The kit is intended for use with adherent cell samples.

CDK7 is a member of the cyclin-dependent kinase family that forms a trimeric complex with cyclin H and MAT1, functioning as a Cdk-activating kinase (CAK) which activates CDK1, -2, -4 and -6. It is an essential component of the transcription factor TFIIH, involved in transcription initiation and DNA repair, and phosphorylates RNA polymerase II to enable transcript elongation. Assessing phosphorylation at threonine 170 of CDK7 is relevant to studies of its activation and function in cell cycle progression and gene transcription regulation.
