Description

From Assay Genie, this in-cell ELISA kit enables qualitative measurement of CDK7 protein expression in cultured cells using a colorimetric detection method. Designed as a convenient, lysate-free, high-throughput assay, it uses an indirect format in which target-specific primary antibodies capture CDK7 while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. The kit is provided in a 96-well microplate format.

Reactive with human and mouse samples and intended for adherent cells, the kit supports a dynamic range of > 5000 cells with detection by colorimetric measurement at 450 nm. Storage is at 4°C for 6 months. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density and adjust for plating differences. The assay is suitable for measuring relative CDK7 levels in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on CDK7 expression.

CDK7 is a member of the cyclin-dependent protein kinase (CDK) family. It forms a trimeric complex with cyclin H and MAT1, functioning as a Cdk-activating kinase (CAK) that activates CDK1, -2, -4, and -6. It is an essential component of the transcription factor TFIIH, linking transcription initiation and DNA repair to cell cycle control, and phosphorylates RNA polymerase II to promote transcriptional elongation. Dysregulation of CDK7 has been linked to diseases including cancer and neurodegenerative disorders, making it a target of interest for therapeutic research.
