Description

From Assay Genie, this cell-based ELISA kit is designed for the qualitative detection of CDK5 protein directly in cultured cells using a colorimetric readout at 450 nm. It works in an indirect ELISA format, in which CDK5 is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colour reaction upon substrate addition. The assay is described as lysate-free, high-throughput and sensitive, allowing researchers to measure relative CDK5 levels in adherent cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on CDK5 expression.

Reactivity is reported for human, mouse and rat samples, with a dynamic range of > 5000 cells per well. The kit includes a monoclonal anti-GAPDH antibody to serve as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step is used to determine cell density so that absorbance values can be adjusted for plating differences. The plate format is a 96-well microplate, with storage at 4°C for 6 months.

CDK5 is a serine/threonine protein kinase of the cyclin-dependent kinase family that, unlike other family members, is activated by p35 (CDK5R1) and p39 rather than by cyclins. It plays a key role in neuronal positioning during brain development, as well as in synaptogenesis, neurotransmission, cell differentiation and migration, and has been implicated in neurodegenerative disease research.
