Description

Assay Genie provides a Cell-Based ELISA kit designed to measure CDK2 protein when phosphorylated at threonine 160. The kit uses a colorimetric detection method at 450 nm and is reactive with Human, Mouse, and Rat samples, with a stated dynamic range of > 5000 Cells. It employs an indirect ELISA format in which CDK2 (Phospho-Thr160) is captured by phospho-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody; the conjugated HRP enzyme then catalyses a colorimetric reaction upon substrate addition.

As a lysate-free, high-throughput assay, the kit supports measurement of the relative amounts of phosphorylated CDK2/CDC2 in cultured cells and can be used to screen the effects of various treatments, inhibitors, or activators on CDK2/CDC2 phosphorylation. The assay is suitable for adherent cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method is provided to determine cell density so that absorbance values can be normalised to cell amounts. CDK2 is described as a key regulator of cell cycle progression, and phosphorylation at threonine 160 is an important post-translational modification that impacts its activity; dysregulation of CDK2 phosphorylation has been implicated in various diseases, including cancer, making it a target of interest for therapeutic intervention.
